Українська: Мікротрубочки є порожніми циліндричними об’єднаннями діаметром ~ 24 нм, які складаються з протофіламентів (утворених з α- та β-тубуліну) та перебувають у стані динамічної нестабільності. Цей стан характеризується стохастичними переходами між фазою “порятунку” (або росту (+)-кінця мікротрубочки) та фазою “катастрофи” (або вкорочення (+)-кінця) та є необхідним для виконання мікротрубочками клітинних функцій. “Порятунок” супроводжується утворенням α-/β-тубулінових димерів та їхнім приєднанням до протофіламентів на (+) кінці; при цьому відбувається зв’язування ГТФ та після вбудовування у решітку гідроліз ГТФ з утворенням ГДФ (відбувається у міждимерному інтерфейсі та є необхідним для динамічної нестабільнсоті). При “катастрофі”, навпаки, відбувається вивільнення субодиниць тубуліну, ГТФ та ГДФ. Мікротрубочки як структурний компонент веретена поділу є необхідними для мітозу, що робить їх важливими мішенями при лікуванні злоякісних новоутворень. Молекули (ліганди), що зв'язуються з різними сайтами α- та β-тубуліну, за характером впливу на мікротрубочки поділяються на стабілізуючі та дестабілізуючі агенти. Відображені на рисунку таксол та пелорузид належать до стабілізуючих агентів, що при зв’язуванні з β-тубуліном викликають укріплення латеральних зв'язків між прилеглими протофіламентами у складі мікротрубочки, запобігаючи “катастрофі” та викликаючи зупинку клітинного циклу.
Для отримання зображень 3-D моделі фрагменту мікротрубочки була використана структура фрагменту мікротрубочки Sus scrofa, визначена за допомогою методу кріоелектронної мікроскопії та депонована у PDB (Kellog et al. (2017); 5SYE). Cтруктура була візуалізована у ChimeraX (https://www.cgl.ucsf.edu/chimerax/), за допомогою якої були отримані зображення. 2-D зображення пелорузиду та таксолу були отримані за допомогою бібліотеки RDKit (https://www.rdkit.org; Python 3.x wrappers). Створення двовимірних схем та об'єднання всіх компонентів ілюстрації здійснювалося у Adobe Illustrator. English: Microtubules are hollow cylindrical assemblies of ~ 24 nm diameter, which are composed of protofilaments (made of α- and β-tubulin) and exist in a state of dynamic instability. This state is characterized by stochastic transitions between “rescue” phase (i.e., growing of a microtubule (+)-end) and catastrophe phase (i.e., shrinkage of a microtubule (+)-end) and is nessesary for a fulfillment of microtubules cellular functions. During a “rescue” phase α-/β-tubulin dimers are created and attached to protofilaments at the microtubule (+)-end, which is accompanied by GTP binding and its hydrolysis to GDP after dimer integration into a microtubule lattice (occurs in inter-dimer interface and is required for a dinamic instability). On the contrary, during a “catastrophe” phase tubulin subunits, GTP, and GDP are released. Microtubules as structural components of mitotic spindle are necessary for a mitosis, which makes them important targets in malignant tumors treatment. Molecules (ligands) that bind to different sites on α- and β-tubulin are divided into groups of stabilizing and destabilizing agents based on their influence on microtubules. Taxol and peloruside, which are represented on the figure, are stabilizing agents: their binding causes an enhancement of lateral contacts between adjacent protofilaments of microtubule preventing a “catastrophe” and leading to cell cycle arrest.
In the illustration the 3-D model of structure of Sus scrofa microtubule fragment determined using the method of cryo-electron microscopy and deposited in PDB was used (Kellog et al. (2017); id: 5SYE). The structure was visualized in ChimeraX to obtain the images (https://www.cgl.ucsf.edu/chimerax/). Images of peloruside and taxol 2-D structures were created using the RDKit library (https://www.rdkit.org; Python 3.x wrappers). The creation of 2-D schemes and combination of all elements were done in Adobe Illustrator. |
